一株产黄曲霉毒素B1的霉菌的分离鉴定及其产毒能力的研究

摘要:本研究利用CA、YES培养基从42个白酒生产体系样品中共分离出76株霉菌,利用超高效液相色谱-质谱联用检测分析确定其中一株菌株G1具有产黄曲霉毒素B1(AFB1)的能力,该菌株来源于白酒存储环境样品。依据G1菌落形态、菌丝和分生孢子形态、ITS、β-Tubulin、 Calmodulin基因序列的系统发育树分析,鉴定该菌株为Aspergillus flavus。进一步研究了环境因素对G1菌株菌落生长和菌株产毒能力的影响。结果表明培养温度和水活度是影响其产毒能力的主要因素。

Screening and identification of a aflatoxin B1 producing strain and its toxin-producing ability
Abstract:42 samples of Chinese liquor production were cultivated on CA and YES media, and screened using the high performance liquid chromatography couple with mass spectrometry (UPLC-MS/MS). One strain which could produce AFB1 was isolated from Chinens liquor storage environment and named as G1. The target strain was identified by the morphological characteristics as well as nucleotide sequence analysis of ITS, β-Tubulin gene and Calmodulin gene, Strain G1 was identified as Aspergillus flavus. The effects of temperature, pH and water activity on the colony morphology and the AFB1 production were studied.The results indicated that temperature and water activity were the main factors influencing AFB1 production of strain G1.
