鸦胆子油对A549肿瘤球和HL60细胞的药效研究

摘要:抗癌药物药效学评价是抗癌药物研究中的重点之一。鸦胆子油(Brucea Javanica Oil)作为苦木科植物提取物,在多种癌症治疗中展现出诱导凋亡、抑制耐药酶及调节免疫功能等作用,其临床制剂已用于协同放化疗治疗。然而,肿瘤异质性、免疫逃逸及肿瘤三维微环境特征可能影响其药效响应机制。本研究通过构建A549肺癌肿瘤球模型模拟体内肿瘤三维微环境,以及利用HL60免疫细胞,评估了鸦胆子油原液及三个分离组分的药效差异。结果显示:在三维肿瘤球体系中,1×10^-4浓度的鸦胆子油原液可以略微提高A549肿瘤球的活性,但1×10^-6-1×10^-5浓度未产生明显影响;当浓度升高到1×10^-3时,A549肿瘤球表现出明显更高的活性,三个分离组分表现出与原液相似的影响,上述结果说明3D培养方式与传统组织培养相比,可能因为肿瘤不同的生长方式削弱药物的作用;对于HL60免疫细胞中,原液和分离组分在处于1×10^-3浓度时均抑制细胞增殖,但原液在1×10^-6浓度呈现促增殖效应,其他组分低浓度处理无显著变化,表明鸦胆子油对于免疫细胞存在浓度依赖的双向调节特性。研究表明,肿瘤球三维微环境可能降低鸦胆子油抗癌疗效,而其对免疫细胞的药效作用受浓度双向调控,这为优化临床上应用鸦胆子油抗癌治疗及联合免疫治疗提供了实验依据。

Pharmacodynamic Study of Brucea Javanica Oil on A549 Tumor Spheroids and HL60 Cells
Abstract:Pharmacodynamic evaluation of anticancer drugs represents a critical focus in oncological research. Brucea Javanica Oil, a botanical extract from the Simaroubaceae family, demonstrates multifunctional antitumor mechanisms including apoptosis induction, suppression of drug-resistant enzymes, and immunomodulatory effects, with its clinical formulations utilized as adjuvant therapy for chemoradiation. However, tumor heterogeneity, immune evasion, and 3D tumor microenvironment characteristics may influence its pharmacodynamic responsiveness. This study constructed an A549 lung cancer spheroid model to simulate the 3D tumor microenvironment in vivo and employed HL60 immune cells to evaluate pharmacodynamic variations among Brucea Javanica Oil crude extract and three separated components. Results revealed that in the 3D spheroid system, the crude extract at 1×10^-4 concentration slightly enhanced A549 spheroid viability, while 1×10^-6 to 1×10^-5 concentrations exhibited no significant effects. At 1×10^-3 concentration, A549 spheroids displayed markedly higher viability, with the three separated components showing similar trends to the crude extract. These findings suggest that the 3D culture system, compared to traditional monolayer cultures, may attenuate drug efficacy due to altered tumor growth patterns. In HL60 immune cells, both the crude extract and separated components inhibited proliferation at 1×10^-3 concentration, whereas the crude extract exhibited pro-proliferative effects at 1×10^-6 concentration, with no significant changes observed for other components at lower concentrations, indicating concentration-dependent bidirectional regulatory properties of the oil on immune cells. The study demonstrates that 3D tumor microenvironments may diminish the anticancer efficacy of Brucea Javanica Oil, while its immunomodulatory effects are bidirectionally regulated by concentration gradients, providing experimental insights for optimizing clinical applications in anticancer therapy and combinatorial immunotherapy strategies.
