工程牛奶外泌体载药系统的制备与抗癌应用

摘要:(1)采用差速离心 法从新鲜牛奶中分离获得高纯度牛奶外泌体,通过化学气相沉积技术合成磁性FeCo@G纳米颗粒,继而运用超声破碎结合膜融合技术成功制备负载FeCo@G的工程牛奶外泌体载药系统(M-FNP)。通过粒径分析、Zeta电位测定及透射电镜形态学观察,确证M-FNP的成功构建。生物相容性评价显示,该工程外泌体载药系统具有优异的细胞相容性。进一步将阿霉素包封于M-FNP中构建药物递送系统(M-DFNP),体外抗癌实验表明,仅2 μg/mL的M-DFNP即可对癌细胞产生显著的毒性作用,展现出优异的癌细胞杀伤效果。

Synthesis and Anticancer Application of Engineered Milk Exosome Drug Delivery System
Abstract:High-purity milk exosomes were isolated from fresh milk using differential centrifugation. Magnetic FeCo@G nanoparticles were synthesized via chemical vapor deposition, followed by the successful preparation of engineered milk exosomes loaded with FeCo@G (M-FNP) through ultrasonic disruption combined with membrane fusion techniques. The successful construction of M-FNP was confirmed by particle size analysis, Zeta potential measurement, and morphological observation via transmission electron microscopy. Biocompatibility evaluation demonstrated that these engineered exosomes exhibited excellent cytocompatibility. Doxorubicin was further encapsulated into M-FNP to construct a drug delivery system (M-DFNP). In vitro anticancer experiments revealed that M-DFNP at a concentration as low as 2 μg/mL exerted significant toxic effects on Synthesis and Anticancer Application of Engineered Milk Exosome-Based Drug Delivery Systemcancer cells, showcasing superior anticancer efficacy.
