多功能小檗碱/黄芩苷自组装药物的制备与表征

摘要:本研究通过一步自组装反应将小檗碱(Berberine, BBR)和黄芩苷(Baicalin, BA)制备成BA-BBR纳米粒子(BA-BBR NPs),并进一步利用咖啡酸和铁离子络合的金属多酚网络(Metal-polyphenol network, MPN)进行修饰,成功合成了新型BA/BBR@MPN纳米颗粒(BA/BBR@MPN NPs)。通过表征分析,BA/BBR@MPN NPs呈球形,且平均粒径为241.15±26.52 nm。在弱酸性环境中,MPN会分解产生铁离子,铁离子发生类芬顿反应,催化过氧化氢产生羟自由基,实现化学动力学治疗。由此产生的羟自由基还会消耗谷胱甘肽水平。光热性能实验结果表明,BA/BBR@MPN NPs升温效果具有浓度依赖性和功率依赖性,且多次激光辐照后依旧保持良好的光热稳定性,光热转换效率为26.4%。细胞毒性实验及体外溶血实验结果表明,BA/BBR@MPN NPs具有良好的生物相容性。综上,本文成功地合成了一种可以实现化学动力学治疗与光热治疗联合作用的多功能BA/BBR@MPN NPs纳米平台,为后续的抗菌或抗肿瘤提供了新的思路。

Preparation and Characterization of multifunctional Berberine/Baicalin self-assembled drugs
Abstract:This study prepared berberine (BBR) and baicalin (BA) into BA-BBR nanoparticles (BA-BBR NPs) through a one-step self-assembly reaction. Subsequently, the nanoparticles were further modified using a metal-polyphenol network (MPN) formed by the coordination of caffeic acid and iron ions, successfully synthesizing novel BA/BBR@MPN nanoparticles (BA/BBR@MPN NPs). Characterization analysis revealed that the BA/BBR@MPN NPs exhibited a spherical morphology with an average particle size of 241.15 ± 26.52 nm. Under weakly acidic conditions, the MPN decomposes to release iron ions, which subsequently trigger a Fenton-like reaction, catalyzing hydrogen peroxide to generate hydroxyl radicals for chemodynamic therapy (CDT). The generated hydroxyl radicals ( OH) further deplete glutathione (GSH) levels. Photothermal performance experiments demonstrated that BA/BBR@MPN NPs exhibit concentration-dependent and power-dependent temperature elevation, maintaining excellent photothermal stability even after multiple laser irradiations, with a photothermal conversion efficiency of 26.4%. Cytotoxicity assays and in vitro hemolysis tests confirmed the excellent biocompatibility of BA/BBR@MPN NPs. In conclusion, this study successfully developed a multifunctional BA/BBR@MPN NPs platform capable of combined chemodynamic therapy (CDT) and photothermal therapy (PTT), providing novel insights for potential antibacterial or antitumor applications.
