柠檬酸转运蛋白SLC25A1在小鼠焦虑样行为中的作用及机制研究

摘要:焦虑症是一种常见的精神类疾病,其发病机制复杂,尚未完全阐明。组蛋白乙酰化作为一种重要的表观遗传修饰,在焦虑、应激等动物模型中已被证实具有重要作用。SLC25A1作为线粒体膜上柠檬酸的转运蛋白,影响乙酰辅酶A的合成,与组蛋白乙酰化修饰过程密切相关。SLC25A1在糖脂代谢中发挥重要作用,但在神经疾病尤其是精神情绪类疾病,如焦虑症中鲜有研究,因此本课题对SLC25A1与焦虑症的作用及机制进行探究。利用腺相关病毒构建在小鼠脑中特异性敲除Slc25a1模型(Slc25a1bKO),发现在皮层和下丘脑等脑区中成功敲除SLC25A1。行为学实验发现在旷场和高架十字迷宫等焦虑行为学范式中,Slc25a1bKO小鼠表现出显著的焦虑样行为,同时伴有睡眠减少。最后通过ELISA检测、WB实验探究其初步分子机制,研究发现Slc25a1bKO小鼠皮层脑区中乙酰辅酶A含量显著降低,H3K27ac水平显著减少。以上结果表明,SLC25A1可能通过调控乙酰辅酶A水平,影响组蛋白H3K27ac修饰,进而调控小鼠的焦虑行为。

The Role and Mechanism of Citrate Carrier SLC25A1 in Anxiety-like Behavior in Mice
Abstract:Anxiety disorder is a common mental illness with a complex pathogenesis that has not been fully elucidated. Histone acetylation, as an important epigenetic modification, has been proven to play a significant role in animal models of anxiety and stress. SLC25A1, as a citrate transporter on the mitochondrial membrane, affects the synthesis of acetyl-CoA and is closely related to the histone acetylation modification process. SLC25A1 plays an important role in glycolipid metabolism, but there are few studies on its role in neurological diseases, especially mental and emotional disorders such as anxiety disorder. Therefore, this study explores the role and mechanism of SLC25A1 in anxiety disorder. By using adeno-associated virus to construct a mouse model with specific knockout of Slc25a1 in the brain (Slc25a1bKO), it was found that SLC25A1 was successfully knocked out in brain regions such as the cortex and hypothalamus. Behavioral experiments showed that Slc25a1bKO mice exhibited significant anxiety-like behaviors in anxiety behavioral paradigms such as the open field and elevated plus maze, accompanied by reduced sleep. Finally, ELISA and WB experiments were used to explore the preliminary molecular mechanism. The results showed that the content of acetyl-CoA in the cortical brain region of Slc25a1bKO mice was significantly reduced, and the level of H3K27ac was significantly decreased. These results suggest that SLC25A1 may regulate anxiety behavior in mice by controlling the level of acetyl-CoA and affecting histone H3K27ac modification.
