摘要:蛋白A(Protein A)亲和层析作为目前最常用的抗体纯化方法,如何提高其使用寿命和降低纯化成本一直在被人们探索。从蛋白A结构域Z出发,基于EpHod模型预测了蛋白A结构域Z单体和四聚体的同源蛋白最适pH分布,筛选了碱性的蛋白A同源序列,在此基础上利用EpHod模型和MPNN模型分析预测了同源突变位点,通过动力学模拟,SDS-PAGE和灰度分析的湿实验验证后,确定了Mu-29-tetra可基本耐受0.5 M NaOH。在0.5 M NaOH中浸泡24 h后仍有80.6%的残余。以免疫球蛋白G(IgG)抗体蛋白作为测试蛋白,测试比对了重组蛋白A亲和层析介质的耐碱性和重复使用性能。0.5 M NaOH浸泡24 h后Mu-29-tetra动态载量保留量分别为90.3%;模拟重复使用100次后动态载量保留量为89.7%。其评价指标基本达到商品化介质水平,具有潜在的市场化应用价值。

Construction of alkali-resistant recombinant protein A based on the EpHod model
Abstract:Protein A (Protein A) affinity chromatography, as the most commonly used antibody purification method, has been explored to improve its longevity and reduce the purification cost. Starting from protein A structural domain Z, we predicted the optimal pH distribution of homologous proteins of protein A structural domain Z monomer and tetramer based on the EpHod model, and screened protein A homologous sequences of basicity, on the basis of which, we predicted the homologous mutation sites using the EpHod model and the MPNN model analysis, and after validation by wet experiments of kinetic simulation, SDS-PAGE and grayscale analysis, we determined that Mu- 29-tetra is largely tolerant to 0.5 M NaOH. 80.6% remains after 24 h immersion in 0.5 M NaOH. The alkaline resistance and reusability of recombinant protein A affinity chromatography media were compared using immunoglobulin G (IgG) antibody protein as the test protein. 90.3% of dynamic load retention was observed after 24 h of immersion in 0.5 M NaOH, and 89.7% of dynamic load retention was observed after simulated reusability for 100 times. The evaluation indexes basically reach the level of commercialized medium, with potential marketable application value.
