着丝粒聚集调控动粒Mis12蛋白复合物的组装

摘要:在有丝分裂间期,裂殖酵母染色体呈现Rabl构型:着丝粒聚集在近纺锤体极体(Spindle Pole Body, SPB)的核膜上,端粒则固定在核膜的另一侧。在聚集的着丝粒上,动粒蛋白稳定存在,在Rabl构型缺陷的突变体中,动粒在间期部分解体,在进入有丝分裂时重新组装。其中动粒蛋白Mis12被认为在间期稳定存在于着丝粒上。本研究通过标记Mis12及Mis12复合物组分Mis13/DSN1,发现其在破坏着丝粒聚集的csi1Δ突变体中,Mis12复合物显著解离。这一结果表明动粒的不同组分对Rabl构型的依赖性存在差异。。

Centromere cluster regulates the assembly of the kinethcore Mis12 complex
Abstract:During the interphase of mitosis, chromosomes in Schizosaccharomyces pombe exhibit a Rabl configuration: centromeres cluster at the nuclear envelope beneath the spindle pole body (SPB), while telomeres are anchored on the opposite side of the nucleus. Meanwhile, kinetochores remain stably assembled on the clustered centromeres. In mutants defective in the Rabl configuration, kinetochores partially disassemble in interphase and reassemble at the onset of mitosis. The kinetochore Mis12 complex is thought to stably localize at the centromere during interphase. In this study, we labeled Mis12 and Mis13/DSN1, another component of the Mis12 complex, with GFP and observed significant dissociation of Mis13, but not Mis12, from centromeres in the csi1Δ mutant, which is defective in centromere clustering. These results indicate that different kinetochore components exhibit varying degrees of dependence on the Rabl configuration.
