中药益生菌复合剂对肥胖相关胰岛素抵抗小鼠的影响研究

摘要:本研究旨在小鼠模型中探究中药益生菌复合剂(Herb medicine-Probiotics Mix,HPM)对肠道菌群及肥胖相关胰岛素抵抗的影响。野生C57BL/6J雄性小鼠随机分为4组,分别是正常饮食组(ND)、高脂饮食组(HFD)、高脂饮食+PBS组(HFD-PBS)、高脂饮食+HPM组(HFD-HPM)。17周后测定小鼠肥胖程度,血糖和胰岛素抵抗水平,分离附睾脂肪组织的免疫细胞进行流式分析,通过16S rDNA测序分析粪便菌群。结果表明,与ND小鼠比较,HFD小鼠体重增加、空腹血糖和胰岛素水平显著升高、葡萄糖和胰岛素耐受异常、胰岛素抵抗增强。HPM干预显著降低高脂小鼠的空腹血糖和胰岛素抵抗。HPM干预小鼠的体重、肥瘦比、血脂相对较低。HFD小鼠eWAT中巨噬细胞和CD8+T细胞显著增多,HPM干预未改变高脂小鼠eWAT中巨噬细胞的数量,但抑制了高脂小鼠CD8+T细胞的增加。HFD小鼠粪便菌群的α多样性增加、β多样性存在差异、拟杆菌门与厚壁菌门的比值降低、Parabacteroides增多及拟杆菌属减少。与HFD-PBS小鼠相比,HPM干预显著提高菌群的乳酸杆菌属丰度、降低Parabacteroides丰度。因此中药益生菌复合剂(HPM)可有效抑制高脂饮食诱导的肥胖相关胰岛素抵抗和代谢紊乱。HPM可能通过调节肠道菌群,减少eWAT中CD8+T细胞数量来抑制胰岛素抵抗。

Effects of herb medicine - probiotics mixes on obesity-induced insulin resistance in mice
Abstract:This study aimed to investigate the effects of Herb medicine-Probiotics Mix (HPM) on gut microbiota and insulin resistance related to obesity in mice. Wild-type C57BL/6J male mice were randomly divided into four groups: normal diet group (ND), high-fat diet group (HFD), high-fat diet + PBS group (HFD-PBS), and high-fat diet + HPM group (HFD-HPM). After 17 weeks, the body weight, blood glucose and insulin levels were detected. Immune cells in epididymal white adipose tissue (eWAT) were examined by flow cytometry, and the fecal microbiome was analyzed by 16S rDNA sequencing. The results showed that compared with the ND mice, the HFD mice showed significantly increased body weight, higher fasting blood glucose and insulin level, impaired glucose and insulin tolerance, worsened insulin resistance. HPM intervention significantly lowered the fasting blood glucose and insulin resistance . The body weight, fat to lean ratio, serum lipid levels were lower in the HPM-treated mice relative to the HFD-PBS mice. HFD mice had significantly higher numbers of macrophages and CD8+T cells in adipose tissue. HPM intervention did not alter adipose tissue macrophage number, but appreciably reduced the number of CD8+. HFD mice showed an increase in α-diversity and difference in β-diversity of the fecal microbiome, with an enhanced abundance of Parabacteroides and decrease in Bacteroides. Compared to HFD-PBS mice, HPM treatment significantly enhanced abundance of Lactobacillus and decreased abundance of Parabacteroides. Therefore, Herb medicine-Probiotics Mix (HPM) can effectively suppress obesity-related insulin resistance and metabolic disorders induced by high fat diet. HPM inhibited insulin resistance possibly by regulating gut microbiome and reducing the number of CD8+T cells in eWAT.
