斑马鱼溶血性贫血模型的建立及枸杞多糖的功效初探

摘要:该文建立了斑马鱼溶血性贫血模型,并利用该模型初步探索枸杞多糖的功效。采用苯肼孵育斑马鱼的方法建立溶血性贫血模型,通过其存活率和心脏红细胞染色情况确立适宜的建模条件,并以其存活率、形态发育、运动行为和组织愈合能力为指标对该模型进行分析,最后初步探索枸杞多糖(polysaccharides from Lycium barbarum,LBP)对斑马鱼溶血性贫血的改善作用。结果表明,斑马鱼溶血性贫血模型的适宜建模条件为2 μmol/L苯肼处理2 dpf(days post fertilization)斑马鱼24 h;与对照组相比,溶血性贫血对斑马鱼的存活率和形态发育没有显著影响,但显著性抑制了其自主运动和组织愈合能力;枸杞多糖处理后,溶血性贫血斑马鱼的心脏红细胞量表现出显著性增加。综上所述,该研究成功建立了斑马鱼溶血性贫血模型,验证了枸杞多糖对该模型具有良好的改善溶血性贫血的功效,为溶血性贫血的研究和枸杞多糖等活性因子的功效探索提供新的实验依据和研究思路。

Establishment of a zebrafish hemolytic anemia model and preliminary exploration of the efficacy of polysaccharides from Lycium barbarum
Abstract:This article established a zebrafish hemolytic anemia model and explored the efficacy of polysaccharides from Lycium barbarum (LBP) using this model. The hemolytic anemia model was established by incubating zebrafish with phenylhydrazine. The suitable conditions were discovered based on the results of survival rate and cardiac red blood cell staining. To further investigate this model, the survival rate, morphological development, motor behavior, and tissue healing ability were analyzed. Finally, the improvement effect of LBP on hemolytic anemia in zebrafish was preliminarily explored. The results showed that the suitable conditions for establishing the zebrafish hemolytic anemia model were treating 2dpf (days post fertilization) zebrafish with 2 μmol/L phenylhydrazine for 24 hours. Compared with the control group, the survival rate and morphological development of zebrafish were not significantly changed in the hemolytic anemia group. However, the abilities of spontaneous movement and tissue healing were significantly inhibited by hemolytic anemia. After treated with LBP, the number of cardiac red blood cell of zebrafish with hemolytic anemia showed a significant increase. In summary, this study established a zebrafish hemolytic anemia model and confirmed the improvement effect of LBP on hemolytic anemia, providing new experimental basis and research ideas for studies on hemolytic anemia and efficacy exploration of active factors such as LBP.
