BMAL1过表达促进结直肠癌Caco-2细胞增殖与迁移的作用研究

摘要:结直肠癌(Colorectal cancer, CRC)是全球范围内的主要恶性肿瘤之一,其发病率和死亡率呈上升趋势。本研究聚焦于脑和肌肉芳基烃受体核转运体样1(Brain and muscle ARNT-like-1,Bmal1),一个在调控生物节律中起关键作用的基因,探讨其在结直肠癌中的潜在作用。通过构建Tet-on诱导表达系统,我们在Caco-2细胞系中实现了BMAL1的过表达,并评估了其对细胞增殖和迁移的影响。实验结果表明,与对照组相比,BMAL1过表达组的细胞增殖率显著提高,EdU+细胞数量增多,细胞周期相关基因表达上调。此外,BMAL1过表达还显著增强了Caco-2细胞的迁移能力,这一点通过划痕实验得到了证实。Western Blot分析进一步揭示了BMAL1过表达可能通过调节Rho相关激酶ROCK1、粘附连接蛋白β-Catenin、钙粘蛋白E-cadherin的表达和粘着斑激酶FAK的磷酸化来促进细胞迁移。本研究的发现为理解BMAL1在结直肠癌发展中的作用提供了新视角。

Study on the Role of BMAL1 Overexpression in Promoting Proliferation and Migration of Caco-2 Cells
Abstract:Colorectal cancer (CRC) is one of the leading malignant tumors globally, with increasing incidence and mortality rates. This study focuses on Brain and Muscle ARNT-like-1 (Bmal1), a key gene in regulating biological rhythms, to explore its potential role in CRC. By constructing a Tet-on inducible expression system, we achieved BMAL1 overexpression in Caco-2 cell lines and assessed its impact on cell proliferation and migration. Experimental results showed that, compared to the control group, BMAL1 overexpression significantly increased cell proliferation rate, EdU+ cell count, and the expression of cell cycle-related genes. Additionally, BMAL1 overexpression markedly enhanced Caco-2 cell migration, as evidenced by scratch assays. Western Blot analysis further revealed that BMAL1 overexpression may promote cell migration by regulating the expression of Rho-associated kinase ROCK1, adhesion protein β-Catenin, cadherin E-cadherin, and the phosphorylation of focal adhesion kinase FAK. These findings provide new insights into the role of BMAL1 in CRC development.
