黄精在九蒸九晒炮制中多糖含量与构成的变化研究

摘要:为了建立一种使用高效液相色谱法测定黄精九蒸九晒炮制的多糖含量与构成变化的方法。根据2020版中国药典中的方法对黄精进行九蒸九晒炮制,运用苯酚-硫酸法测定九蒸九制黄精炮制过程中的多糖含量变化。使用三氟乙酸(TFA)水解多糖,1-苯基-3-甲基-5-吡唑啉酮(PMP)用于单糖的柱前衍生化。实验结果显示从生黄精样品到九蒸九晒黄精,四蒸四晒黄精的多糖含量最高。黄精多糖由八种单糖构成,分别是D-甘露糖、D-核糖、L-鼠李糖、D-葡萄糖醛酸、D-葡萄糖、D-半乳糖、D-阿拉伯糖、L-岩藻糖。对黄精的九蒸九晒炮制不影响黄精多糖的单糖构成,但会使不同单糖的含量发生改变。

Research on the changes of polysaccharide content and monosaccharide composition of Polygonatumsibiricum in the processing of nine steaming and nine shining
Abstract:ITo establish a method for the determination of polysaccharide content and composition changes of Polygonatum sibiricum in the processing of nine steaming and nine shining processed by high performance liquid chromatography. According to the method in the 2020 edition of the Chinese Pharmacopoeia, Polygonatum sibiricum was processed with nine steaming and nine shining, and the phenol-sulfuric acid method was used to determine the changes in polysaResearch on the changes of polysaccharide content and monosaccharide composition of Polygonatum sibiricum in the processing of nine steaming and nine shiningccharide content during the processing of Polygonatum sibiricum with nine steaming and nine shining. Trifluoroacetic acid (TFA) was used for hydrolyzing polysaccharides and 1-phenyl-3-methyl-5-pyrazolinone (PMP) was use d for pre-column derivatization of monosaccharides. The results of experiment showed that the polysaccharide content of four steamed and four shined polygonatum sibiricum was the highest from the sample of polygonatum sibiricum to the nine steamed and nine shined polygonatum sibiricum samples. Polygonatum sibiricum polysaccharide is composed of eight monosaccharides: D-mannose, D-ribose, L-rhamnose, D-glucuronic acid, D-glucose, D-galactose, D-arabinose, and L-fucose. Nine steaming and nine shining processing did not change the monosaccharide composition of polygonatum sibiricum polysaccharides, but it changed the content of different monosaccharides.
