紫丁香蘑液体发酵优化以及其水提、碱提多糖的结构与功能活性研究

摘要:紫丁香蘑是一种名贵的药食用野生真菌,其具有非常高的抗氧化、抗癌、抗病毒等活性。本研究对紫丁香蘑的液体发酵培养基进行单因素以及正交优化,最佳发酵培养基为3%(w/v)葡萄糖、1%酵母粉、0.05%磷酸二氢钾、0.25%硫酸镁,此条件下的紫丁香蘑生物量为22.3g/L。从紫丁香蘑菌丝体中用热水提取法和热碱提取法提取了水提紫丁香蘑多糖(WLNP),碱提紫丁香蘑多糖(ALNP),用FT-IR、HPGPC、离子色谱法、SEM对多糖的结构进行了结构表征,结果表明WLNP(1532kDa)分子量比ALNP(767kDa)大,扫描电镜下WLNP较为规则而ALNP呈大块的片状。WLNP主要由GlcN、Glc、Man组成, ALNP主要由Glc、Man、Fru组成,主要优势单糖都是Glc。在抗氧化活性研究中,WLNP比ALNP表现出更强大的抗氧化活性,最高DPPH自由基清除率为88.34±2.47%(WLNP)。细胞实验结果显示,WLNP对Hep-G2的抑制率显著高于ALNP, WLNP的IC50(317.9 μg/mL)最低,即表示抗癌活性最强。基于以上的研究,紫丁香蘑多糖有望成为功能食品中新的一员, WLNP在功能食品或药物方面更具有发展的潜力。

Study on the optimization of liquid fermentation of Lepista Nuda and the structure and functional activity of its alkaline- and water-extracted polysaccharides
Abstract:Lepista Nuda is a kind of rare edible wild fungus, which has very high antioxidant, anticancer and antiviral activities. In this study, the liquid fermentation medium of Lepista Nuda was optimized by single factor and orthogonal optimization. The optimal fermentation medium was 3% (w/v) glucose, 1% yeast powder, 0.05% potassium dihydrogen phosphate and 0.25% magnesium sulfate. The biomass of Lepista Nuda under this condition was 22.3g/L. Water-extracted polysaccharide (WLNP) and alkali-extracted polysaccharide (ALNP) were extracted from the mycelium of Lepista Nuda by hot water and hot alkali. The structure of polysaccharides was characterized by FT-IR, HPGPC, ion chromatography and SEM. The results showed that WLNP (1532kDa) was larger than ALNP (767kDa), and WLNP was more regular under scanning electron microscope, while ALNP was a large flake. WLNP is mainly composed of GlcN, Glc and Man, ALNP is mainly composed of Glc, Man and Fru, and Glc is the main dominant monosaccharide. In the study of antioxidant activity, WLNP showed more powerful antioxidant activity than ALNP, and the highest DPPH free radical clearance was 88.34±2.47%(WLNP). The results of cell experiments showed that WLNP had a significantly higher inhibition rate on Hep-G2 than ALNP, and WLNP had the lowest IC50 (317.9 μg/mL), indicating the strongest anticancer activity. Based on the above research, polysaccharides from Lepista Nuda are expected to become a new member of functional food, WLNP has more development potential in functional food or medicine industry.
