摘要:漆酶是一种含铜的多酚氧化酶,属于蓝色多铜氧化酶家族,能够氧化,多种芳香化合物和其它有机化合物,并使氧气还原生成水。漆酶催化的底物种类广泛,因此在纸张漂白、生物修复、染料脱色、果汁与酒的澄清和纺织品的清洁等方面具有巨大的应用潜力。漆酶在大肠杆菌异源表达时易形成包涵体,可溶性蛋白表达量很低。本文通过漆酶与锚定肽(GDGDD)融合并与分子伴侣素 MA4386 共表达的方法来提高目的蛋白在大肠杆菌中的可溶性表达,最终获得了具有生物活性的高纯度漆酶。?????

Prokaryotic expression, purification and characterization of laccase from the fungus Cyathus bulleri
Abstract:Laccase is a copper-containing polyphenol oxidase, belonging to the blue polycopper oxidase family, capable of oxidizing, a wide range of aromatic compounds and other organic compounds and reducing oxygen to form water. Laccase catalyzes a wide range of substrates, and thus has great potential for applications in paper bleaching, bioremediation, dye decolorization, clarification of fruit juices and wines, and textile cleaning. Laccase is prone to form inclusion bodies when heterologously expressed in Escherichia coli, and soluble protein expression is very low. In this paper, the fusion of laccase with an anchoring peptide (GDGDD) and co-expression with the molecular chaperonin MA4386 were used to improve the soluble expression of the target protein in E. coli, and finally a high-purity laccase with biological activity was obtained.
