aGVHD对小鼠肠道血管形态与结构的影响及其机制

摘要:为了探究aGVHD发病进程对小鼠肠道组织血管的变化和肠道免疫稳态之间的关系和机制。我们建立小鼠aGVHD模型,分析小鼠小肠绒毛血管内皮细胞和周细胞以及浸润免疫细胞在不同时间点的动态变化,并运用qPCR、Smart-seq技术分析寻找差异表达基因。发现在aGVHD的发病进程中,小肠绒毛血管的损伤及浸润免疫细胞的变化与疾病进程密切相关,与炎症反应、细胞凋亡及肠道屏障相关基因表达显著变化。根据研究结果,我们认为aGVHD诱发的小肠绒毛血管的严重损伤和结构紊乱可能是导致受体死亡的一个重要原因,小肠绒毛的aGVHD炎症反应可能主要通过影响与内皮细胞凋亡及肠道屏障相关的基因表达,从而破坏小肠绒毛血管的完整性和提高其通透性。

The effects of aGVHD on the morphology and structure of murine intestinal blood vessels and its underlying mechanism
Abstract:To explore the relationship and mechanism between the changes of intestinal tissue blood vessels and intestinal immune homeostasis in mice during the course of aGVHD. We established a mouse aGVHD model, and analyzed the dynamic changes of endothelial cells, pericyte and infiltrating immune cells in mouse small intestinal villi at different time points. qPCR and Smart-seq were used to search for differentially expressed genes. It was found that in the course of aGVHD, the damage of small intestinal villus vessels and the changes of infiltrating immune cells were closely related to the course of the disease, and the expression of inflammatory response, apoptosis and intestinal barrier related genes were significantly changed. According to the results of the study, we believe that the severe damage and structural disorder of villi vessels induced by aGVHD may be an important cause of receptor death, and the aGVHD inflammatory response of villi may damage the integrity of villi vessels and improve their permeability mainly by affecting gene expression related to endothelial cell apoptosis and intestinal barrier.
