中药连花清瘟颗粒的胃肠道消化吸收过程研究

摘要:体外消化吸收模型可用于研究中药制剂在代谢前的释放、转运和吸收。它对药物剂型开发、联合用药和药理机制研究具有重要价值。中药制剂的药效是由各个成分通过协同作用产生的,因此探讨制剂中多种成分下的消化吸收以及它们之间可能的相互作用具有深刻意义。本文选择了一种抗病毒中药制剂-连花清瘟颗粒作为研究对象。结合LC-MS,通过体外消化模型和Ussing chamber吸收模型模拟了连花清瘟颗粒的消化和吸收过程。体外消化实验结果显示,主要成分的表现各有不同,连翘苷基本没有变化,而连翘酯苷A的降解程度最高。进一步考察了颗粒中连翘酯苷A和连翘酯苷E在消化过程中的关系,以及初步鉴定了主要成分的消化降解产物,这很可能与连花清瘟的生物利用度和疗效有关。在模拟肠道吸收实验中,统计了主要成分的表观渗透系数、肠道吸收率和口服利用度。这项研究有助于了解连花清瘟颗粒在体内的变化,明确多种成分的吸收效果,更好地促进其有效使用。

Investigation of the gastrointestinal digestion and absorption process of Chinese medicine LianhuaQingwen granules
Abstract:In-vitro digestion and absorption models can be used to study the release, transport and absorption of Chinese herbal preparations before metabolism. It is valuable for dosage formulation development, drug combination and mechanism researches. The efficacy of Chinese herbal preparations is produced by the individual components through synergistic effects, and it is deeply significant to explore the digestion and absorption under multiple components together with the possible interaction effects between them.Investigation of the gastrointestinal digestion and absorption process of Chinese medicine LianhuaQingwen granules In this paper, an antiviral Chinese herbal preparation, LianhuaQingwen (LHQW) granules, was chosed as the subject. Combined with LC-MS, the digestion and absorption process of LHQW granules was revealed by in vitro digestion model and the Ussing chamber. The results showed that the transformation of the main indicative ingredients manifested differently, and phillyrin remained largely unchanged, while forsythoside A was the most degraded. The relationship between forsythoside A and forsythoside E during digestion process was examined firstly, which may well be related to the bioavailability and efficacy of this herbal treatment. The digestive degradation of some major components was also evaluated. By simulating intestinal absorption through the Ussing chamber, the apparent permeability coefficient, intestinal penetration rate and oral availability of major components were investigated. This study will contribute to understand the changes of LHQW granules in the body and clarify the absorption effect in the co-existence of multiple ingredients, which can promote its effective use.
