生物材料-聚羟基脂肪酸酯的合成研究进展

摘要:聚羟基脂肪酸酯(Polyhydroxyalkanoates, PHAs)-一类微生物在碳氮失衡的情况下合成的脂肪族聚酯,因为具有优异的生物降解性、压电性以及生物相容性等多种材料学性能,除了作为一类可替代传统塑料的包装材料之外,在医疗领域的应用也广受关注。然而,由微生物发酵生产的PHAs,因为合成成本较高和合成聚合物的结构单一等问题导致其应用推广困难。因此,降低PHAs的成本和新型PHAs的开发成为目前的研究热点。本文针对PHAs合成存在的问题,从代谢工学角度综述利用PHAs代谢途径改造增加收率方面研究结果。另外从生物合成角度,结合本研究团队利用体外途径研发的新型PHAs工作,综述目前新型PHAs合成研究结果。并对PHAs合成存在的问题给出解决方案,为其应用推广以及新型产物研究提供借鉴。

Biomaterial-Progress in synthesis of Polyhydroxyalkanoates esters
Abstract:Polyhydroxyalkanoates ( PHAs ) are a kind of aliphatic polyesters synthesized by microorganisms under the condition of carbon and nitrogen imbalance. Due to their excellent biodegradability, piezoelectricity and biocompatibility, PHAs have attracted wide attention in the medical field as well as a kind of packaging materials that can replace traditional plastics.However, PHAs produced by microbial fermentation are difficult to be widely used because of high synthesis cost and single structure of synthetic polymers.Therefore, reducing the cost of PHAs and developing new PHAs have become current research hotspots. In this paper, aiming at the problems of PHAs synthesis, the research results of PHAs metabolic pathway modification to increase yield are reviewed from the perspective of metabolic engineering.In addition, from the perspective of biosynthesis, combined with the new PHAs work developed by our team in vitro, the current research results of new PHAs synthesis were reviewed. The solutions to the problems in PHAs synthesis were also given, which provided reference for its application and promotion and new product research.
