松材线虫接种培养条件对其生长繁殖的影响

摘要:从培养基浓度、接种数量、保存时间、接种方法考察对松材线虫的培养效果。结果表明,接种初期,除直接接种线虫的处理外,从培养基中分离出的线虫数量会少于接种量。松材线虫在低浓度培养基上也可生长,但繁殖数量少。接种数量较少的处理,线虫在培养基中繁殖时间加长。接种量较大的处理,虽然线虫快速繁殖,实验时间变短,但有大量线虫爬行至培养皿盖上,线虫归集不便也会造成计数误差。在各个保存时期,新鲜(刚培养出)松材线虫的活力最好,将线虫从原有培养基上直接接种入新的培养基线虫繁殖快数量多。材线虫在培养基上的繁殖速度和数量与培养基浓度、接种量、保存时间和保存方法等多个因素有关,但并非严格一一对应的关系,在多个因素组合中出现线虫数量的爆发性增长。

Effects of Inoculation and Culture Conditions on the Growth and Reproduction of PWN
Abstract:The culture effect of pine wood nematode (Bursaphelenchus xylophilus,PWN) was investigated from the concentration of culture medium, inoculation quantity, preservation time and inoculation method. The results showed that in the early stage of inoculation, the number of nematodes isolated from the medium was less than the inoculation amount, except for the treatment of direct inoculation. PWN can also grow on low concentration medium, but the number of reproduction is small. The time of nematode breeding in the medium was longer when the inoculation quantity was less. In the treatment with large inoculation amount, the nematodes propagated rapidly and the experimental time became shorter, and a large number of nematodes crawled to the cover of the culture dish which caused the counting error and the collection of nematodes was inconvenient. In each preservation period, fresh (just cultured) PWN had the best vitality. When the nematodes were directly inoculated into the new culture medium from the original culture medium, the population of the nematodes was fast and large. The reproduction speed and quantity of wood nematode on the medium are related to many factors, such as medium concentration, inoculation amount, storage time and storage method, but they are not strictly one-to-one correspondence.
