三液相提取水溶性成分和脂溶性成分

摘要:丹参具有抗动脉粥样硬化、抗神经炎症、抗肿瘤等药理作用。水溶性丹酚酸B(LAB)和脂溶性丹参酮IIA(Tan IIA)是丹参的主要活性成分,传统提取LAB和Tan IIA的工业方法存在成本高、效率低、操作繁琐等问题。本文采用正己烷-乙醇-NaH2PO4-H2O 三液相(TLPS)同时提取LAB和Tan IIA。最佳提取条件为:NaH2PO4溶液(2.50mol/L,pH=2.00):乙醇:正己烷=38:38:24(v/v/v),提取温度为47.0℃,时间为12.00小时。LAB和Tan IIA的总产率为146±9.8mg/g和0.538±0.027mg/g,比药典的单相方法分别提高了128%和52.8%。并且对LAB和Tan IIA的提取和分离过程中的热力学和动力学进行分析讨论。

Extraction of water-soluble and fat-soluble components with three liquid phase system
Abstract:Water-soluble lithospermic acid B (LAB) and fat-soluble tanshinone IIA (Tan IIA) are main bioactive components in Salvia miltiorrhiza Bunge which has anti-atherosclerosis, anti-neuritis, anti-tumor and other pharmacological effects. Traditional industrial methods for extracting LAB and Tan IIA have the problems of high costs, low efficiency and cumbersome operation. In this paper, n-hexane-ethanol-NaH2PO4-H2O three liquid phase system (TLPS) was used to extract LAB and Tan IIA simultaneously. TLPS extraction was optimized to achieve optimal condition: aqueous NaH2PO4 solution (2.50mol/L, pH 2.00): ethanol: n-hexane=38:38:24 (v/v/v), temperature 47.0?C and time 12.00 h. The total yields of LAB and Tan IIA were 146±9.8 mg/g and 0.538±0.027 mg/g, which were 128% and 52.8% higher than the single phase methods in the pharmacopoeia, respectively. The thermodynamics and kinetics in the extraction and separation of LAB and Tan IIA were analyzed and discussed.
