K599诱导中棉所24毛状根转化体系的建立

摘要:[目的]为棉花转基因研究的提升和改进提供方法和奠定基础。[方法]利用发根农杆菌 K599分别侵染棉花中棉所24的子叶、茎段和带子叶节的下胚轴,诱导毛状根产生;利用 PCR技术和GUS组织化学染色对转基因毛状根进行分子和生化检测。[结果]中24子叶和带子叶节的下胚轴在发根农杆菌 K599侵染后均能够产生毛状根,但带子叶节的下胚轴诱导率较高,可以达到66.8%,更适合作为转化外植体。PCR检测和GUS染色表明转基因毛状根扩增有目的条带,染色显著变蓝,为阳性毛状根。[结论]建立并优化了发根农杆菌K599介导的棉花遗传转化体系,成功获得阳性转基因毛状根。

Establishment of cottonZhong24 hairy root transformation system
Abstract:I[Objective] Providing a method and laying a foundation for transgenic study of cotton.[Methods] hairy roots were induced by Agrobacterium tumefaciens k599 infecting the cotyledons, stem segments and hypocotyls of cotyledons and scallop nodes of cotton, and hairy roots were detected by PCR and GUS histochemical staining. [Results] Hypocotyl of cotyledons and scallop nodes could produce hairy roots after infection by A. tumefaciens k599, but Hypocotyl induction rate of cotyledon and scallops was 70%, which was more suitable explants for transformation. PCR and GUS staining showed that the positive PCR bands were amplified and the roots became blue after staining, indicating the successful transformation for hairy roots .[Conclusion] A cotton genetic transformation system mediated by A. tumefaciens K 599 was established and optimized.
