桂花OfMYB1和OfMYB2基因的克隆与表达特性分析

摘要:桂花(Osmanthus fragrance)属于木犀科木犀属植物,具有非常重要的园林应用价值和经济价值。研究利用转录组测序获得的表达序列信息,结合RACE技术,从'镉橙丹桂'和'柳叶金桂'两个桂花品种中筛选并克隆得到两个MYB家族基因。其ORF全长分别为588bp和915bp,各自编码195和304个氨基酸,分别命名为OfMYB1和OfMYB2。序列分析与系统进化树表明,OfMYB1具有明显的R2R3-MYB结构域,与金鱼草中AmMYB305同源性最高,其C端具有保守的nyWsV/ME/DDlWP/S基团。OfMYB2具有R1-MYB结构域,与马铃薯(Solanum tuberosum)中StMYB1R-1同源性最高,同属于CCA1-like Ⅱ亚家族。亚细胞定位实验表明两个基因都定位在细胞核。Real-time PCR比较分析OfMYB1和OfMYB2基因的时空表达特性,结果表明,OfMYB1在花瓣中表达量最高,在花梗、雌蕊中的表达量次之,在叶片中几乎不表达,并在连续3天昼夜时间的表达量具有明显节律,表现为先升高后降低,在中午12:00达到表达高峰。OfMYB2在花瓣和叶片中均有表达,在不同品种不同花期中的表达量均有差异,具有一定的昼夜变化规律。

Cloning and Characterization of OfMYB1 and OfMYB2 genes from Osmanthus fragrance
Abstract: Osmanthus fragrance, belonging to the Oleaceae family, is a well-known ornamental and economically important, aromatic woody plant with a long history and is one of the ten most famous traditional plant in China. With the expressed sequence tags from transcriptome assembly and RACE method, two MYB transcription factors , which may involving in the biosynthesis of aroma and flower pigmentation, were obtained from petals of Osmanthus fragrans. The ORF of these two MYB genes were separately 588bp and 915bp, encoded 195 and 304 amino acid and named OfMYB1 and OfMYB2. Sequence and phylogenetic analysis showed that, OfMYB1 has an obvious R2R3-MYB domain, a highest homology with snapdragon AmMYB305, and a conserved motif nyWsV/ME/DDlWP/S in C-terminal. OfMYB2 has a R1-MYB domain and a highest homology with StMYB1R-1 from potato (Solanum tuberosum), belonging to the CCA1-likeⅡ subfamily. Experiment showed that the two MYB gene were located in nucleus. The results of time and space expression patterns by real-time PCR showed that OfMYB1 had the highest amount of expression in the petals, while no expression in young leaves, comparing to peduncles, pistils and stamens. Besides, it's showed a significant circadian rhythm, reaching the peak expression at 12:00. OfMYB2 were expressed in different expression levels in different varieties of flowering stages and in different tissues, also having a certain circadian variation.
