SERCA家族的功能异常及其在相关疾病中的研究进展

摘要:肌浆(内质)网钙离子ATPase酶(SERCA)的主要功能是通过催化ATP水解将细胞质中的钙离子转运到肌浆(内质)网内,它在控制钙离子信号通路中发挥着关键作用,影响着细胞的生长、凋亡以及一些疾病的形成。近期的研究中明确了SERCA家族在包括肿瘤、心脏病、皮肤病、哮喘等一些疾病中的表达水平的变化与疾病的发生、发展有密切联系。为探究SERCA家族作为疾病发生的重要因素和治疗靶点的提供可能性,本文对SERCA家族的生物学特性、功能异常及其在疾病中的相关研究进行了总结,认为SERCA家族的表达异常直接影响细胞质和内质网中钙离子的稳态,从而影响细胞的恶性增殖、皮肤角质形成和心脏血管等疾病的发生发展。

Abstract:Sarcoplasmic reticulum Ca2+-ATPase (SERCA) is a representative member of P-type ion-transporting ATPases and catalyzes Ca2+ transport coupled with ATP hydrolysis from the cytosol to the sarco(endo)plasmic reticulum, it play a critical role in stimulating cell growth,induing apoptosis and leading to related illness. Recent studies have identified the variational expression level of SERCA family were strongly coupled with development of the cancer, heart disease,dermatosis and asthma. To investigate the effect and possible treatment target for disease development, we summaried the features and studies on SERCA family. It is believed that alterative expression of SERCA damage the calcium homeostasis between cytosol and sarco(endo)plasmic reticulum,resulting in malignant proliferation, keratinocyte formation and occurrence of cardiovascular disease.
