微球固定D-阿洛酮糖3-差向异构酶法合成稀少糖D-阿洛酮糖

摘要:D-阿洛酮糖3-差向异构酶(DPE)是一种能催化D-果糖转化为D-阿洛酮糖的异构酶。本实验采用疏水性介孔聚苯乙烯微球(PST)和新型超大孔聚甲基丙烯酸缩水甘油酯微球(PGMA)偶联D-阿洛酮糖3-差向异构酶,考察了固定化酶的固定条件和酶学性质。实验结果表明,PST和PGMA在水相中的DPE最大负载量分别可以达到40 mg/g和106 mg/g。两种固定化酶的最大转化率分别为28%和29.8%,这与游离酶的最大转化率相差不大。同游离酶相比,固定化酶具有更高的稳定性以及较宽的pH范围。然而载体PST固定化酶的最适温度和最适pH有所改变,变为50 ℃和6.5。PST和PGMA微球固定化酶重复使用性结果表明,在进行5次重复后的相对酶活分别可以达到45%和70%,可以看出PGMA微球固定化酶具有更优的稳定性。

Immobilization of D-psicose 3-epimerase on microspheres for synthesis of rare sugar D-psicose
Abstract:Abstract: D-Psicose 3-epimerase ( DPE) is an enzyme that catalyzes the reversible isomerization between D-fructose and D-psicose. In this study,the GigaPST and GigaPGMA were used as carriers for the immobilization of the D-psicose 3-epimerase. The immobilized conditions and enzymatic properties of the immobilized enzymes were studied.The maximum of loading enzyme amounts on GigaPST and GigaPGMA in aqueous phase reached 40 mg/g and 106 mg/g. The maximum conversionrates of the two immobilized enzymes were 28% and 29.8% with no big difference to free enzyme. Compared with free enzyme, the immobilized DPE showed much higher tolerance for pH and temperature.The optimal temperature and pH of the immobilized enzyme on GigaPST increased to 50℃ and 6.5, respectively. The study of the reusability of DPE on GigaPST and GigaPGMA showed that immobilized enzymes retained 27% and 42% initial activity after 5 cycles, which demonstrated that the property of immobilized enzyme on GigaPGMA was more stable.
