TWIST-2抑制慢性髓系白血病细胞株MEG-01的体外生长

摘要:目的 研究TWIST-2对慢性髓系白血病细胞株MEG-01生长的影响及其机制。 方法 借助慢病毒体系在MEG-01细胞中过表达TWIST-2,检测其对MEG-01细胞增殖、集落生成实验和细胞周期的影响,并通过Western检测TWIST-2对BCR-ABL的表达及下游相关信号通路的影响。 结果 过表达TWIST-2后,MEG-01细胞生长受抑制,集落生成能力显著降低,细胞阻滞在G0/G1期;Western显示相较对照细胞TWIST-2显著抑制BCR-ABL的表达、活性及其下游JAK2-STAT5的活性。结论 TWIST-2可以抑制MEG-01细胞中BCR-ABL的表达和活性,从而抑制其下游JAK2-STAT5信号通路,最终减缓细胞周期进程、抑制细胞的体外生长。

TWIST-2 inhibits the growth of MEG-01 cells in vitro
Abstract:Objective. To study the effects of TWIST-2 on MEG-01 cells (CML cells) and its underlying mechanisms. Methods. Using lentivirus system to deliver TWIST-2 into MEG-01 cells, then proliferation assay, colony-forming cell assay and cell cycle analysis were used to evaluate the effect of TWIST-2 overexpression in MEG-01 cells. Western blot was to used to analyze the expression of BCR-ABL and its downstream components upon TWIST-2 overexpression. Result. Upon TWIST-2 overexpression, the growth of MEG-01 cells was restrained, CFC ability was decreased and cell cycle was arrested in G0/G1 stage. The expression and activity of BCR-ABL was decreased, in addition JAK2-STAT5 signal was attenuated. Conclusion. TWIST-2 is able to inhibit the growth of of MEG-01 cells possibly through the suppression of BCR-ABL/JAK2/STAT5 signaling.
