鬃狼线粒体基因组测序及其系统进化分析

摘要:通过PCR扩增测得鬃狼(Chrysocyon brachyurus)线粒体基因组序列,分析其结构组成,蛋白质编码基因密码子使用情况等,结合已有犬科动物线粒体基因组数据,运用最大简约法,最大似然法和贝叶斯分析法进行系统发育分析。结果表明,鬃狼线粒体基因组全长为16, 760bp,共编码37个基因,包括12S rRNA基因、16S rRNA基因、22个tRNA基因和13个蛋白质编码基因,具有明显的A+T碱基(61.7%)偏好性。蛋白质编码基因在密码子使用上遵循犬科动物的密码子普遍使用规律。与其它犬科动物相比,在tRNA的相同位置,共发现7处一致的碱基错配。系统发育分析表明,犬科动物总体分为两大支系,其中鬃狼与豺属、犬属位于一支。

Mitochondrial genome sequencing and phylogenetic analysis of Chrysocyon brachyurus
Abstract:The complete mitochondrial genome of Chrysocyon brachyurus was firstly sequenced by polymerase chain reaction(PCR), and the structure, composition and codon usage were analyzed. In order to have an insight into the classification and phylogenetic relationships of canids, phylogenetic analyses by maximum parsimony, maximum likelihood, and Bayesian methods were executed respectively. The complete mitochondrial genome sequence of Chrysocyon brachyurus is 16,760 bp long, which containing 12S rRNA gene, 16S rRNA gene, 22 tRNA genes, 13 protein-coding genes and 1 control region. A strong A-T bias(61.7%) is found in mitochondrial genome of Chrysocyon brachyurus. The codon usage of the genome protein-coding genes is in accordance with other canids and 7 unmatched base pairs are detected at the same locations in all canids tRNA genes. The phylogenetic analysis showed that the canids divided into two clades, and Chysocyon, Cuon and Canis belonged to the same one.
