组胺对星形胶质细胞Egr-1mRNA表达的调节作用

摘要:目的:探讨组胺对脑内星形胶质细胞Egr-1 mRNA表达是否具有调节作用。方法:将野生型(WT)小鼠和组氨酸脱羧酶敲除(HDC-KO)小鼠各4只取脑,并提取皮层组织总mRNA。将原代培养的大鼠皮层星形胶质细胞给予不同浓度的组胺(10-8、10-7、10-6、10-5或10-4 M)处理15、30、60、120或240 min。H1受体拮抗剂吡拉明和苯海拉明及H2受体拮抗剂西咪替丁和左拉替丁分别于组胺给药前15 min加入。组胺处理完毕后,提取细胞总mRNA。利用实时定量PCR测定组织及细胞样本的Egr-1 mRNA表达量。结果:与WT小鼠相比,HDC-KO小鼠大脑皮层Egr-1 mRNA表达量显著降低。在培养的星形胶质细胞上,组胺可浓度及时间依赖地促进Egr-1 mRNA表达,其中组胺在10-5 M的浓度下作用即达到最强。此外,组胺(10-5 M)处理30 min时Egr-1 mRNA表达量达到峰值,60 min后降至基础水平,该作用可被组胺H1受体拮抗剂吡拉明和苯海拉明显著抑制,而H2受体拮抗剂西咪替丁和左拉替丁对Egr-1 mRNA表达无明显影响。结论:组胺对大脑皮层及培养的星形胶质细胞Egr-1 mRNA表达具有上调作用,该作用可能与激动组胺H1受体有关。

Effects of histamine on EGR-1 mRNA expression in astrocytes
Abstract:AIM: To explore whether histamine can regulate the expression of Egr-1 mRNA in cerebral cortex astrocytes. METHODS: Normal wild-type (WT) mice (n = 4) and histidine decarboxylase knockout (HDC - KO) mice (n = 4) were sacrificed for extracting the total mRNA of the cerebral cortex. Primary cultured rat cortical astrocytes were treated with 10-8, 10-7, 10-6, 10-5 or 10-4 M histamine for 15, 30, 60, 120 or 240 min. H1 receptor antagonist pyrilamine and diphenhydramine and H2 receptor antagonist cimetidine and zolatidine were pretreated for 15 min followed by 10-5 M histamine exposure for 30 min. After histamine treatment, cell total mRNA was extracted. Using real-time quantitative PCR, the expression of Egr-1 mRNA in tissue and cells was tested. RESULTS: Egr-1 mRNA expression in cerebral cortex of HDC - KO mice was significantly lower than that of WT mice. In cultured astrocytes, histamine induced Egr-1 mRNA expression in a dose-and time-dependent manner. The maximum increase in the level of Egr-1 mRNA was produced by a histamine concentration of 10-5 M. In addition, histamine-induced Egr-1 mRNA accumulation peaked at 30 min and declined to the basal level 60 min after commencing stimulation. Furthermore, histamine-induced Egr-1 mRNA expression was inhibited by H1 receptor antagonist pyrilamine and diphenhydramine but not H2 receptor antagonist cimetidine and zolatidine. CONCLUSION: Histamine can upregulate the Egr-1 mRNA expression in cerebral cortex and cultured astrocytes, which may attribute to H1 receptor activation.
