超声辅助溶剂提取烟草多酚的方法研究*

摘要:通过超声溶剂萃取法提取烟草多酚,用超高效液相色谱(UPLC)进行多酚含量测定,以多酚含量为指标,采用单因素试验及正交试验设计,从溶剂浓度、料液比、超声温度、超声时间等方面来研究烟草多酚最佳提取体系。经分析得出三组烟草多酚最佳提取体系为:60%甲醇,料液比为1∶15,超声温度为50℃,超声时间为30min时,烟草多酚提取量为(27.85±0.20)mg/g;60%乙醇,料液比为1∶10,超声温度为50℃,超声时间为30min时,烟草多酚提取量为(22.73±0.33)mg/g;丙酮浓度为60%,料液比为1∶10,超声温度为40℃,超声时间为10min时,烟草多酚提取量为(25.64±0.82)mg/g。

Study on the extraction of polyphenols from tobacco leaves using ultrasonic-assisted solvent extraction*
Abstract:In this paper, polyphenols were extracted from tobacco leaves with ultrasonic-assistant solvent extraction and the content of polyphenols was determined by ultra-high performance liquid chromatography (UPLC).Both single factor experiment and orthogonal experiment were performed to select the best extraction condition from solvent concentration, material to liquid ratio, temperature and time of ultrasonication. The optimum extraction condition of the three solvent were as follows: 60% methanol, material to liquid ratio of 1∶15, ultrasonic temperature of 50 ℃for 30 min, the content is (27.85±0.20) mg/g; 60% ethanol, material to liquid ratio of 1∶10, ultrasonic temperature of 50 ℃for 30 min, the content is (22.73±0.33) mg/g; 60% acetone, material to liquid ratio of 1∶10, ultrasonic temperature of 40 ℃for 10 min, the content is (25.64±0.82) mg/g.
