壳聚糖/低分子量聚乙烯亚胺复合物载BMP2基因促进骨再生的研究

摘要:通过壳聚糖(CS)和低分子量聚乙烯亚胺(PEI)的接枝共聚反应,我们获得了一种低毒高效的聚合物基因载体,并利用其负载骨形态发生蛋白2基因进行促骨再生的研究。利用动态光散射测得CS-PEI/DNA 复合物的粒度为162nm,电势为26.7mV,聚合物结合基因的能力随着质量比的增加而增强,CS-PEI聚合物在293T和MC3T3-E1两种细胞系中均表现出良好的细胞相容性。通过流式细胞术检测,在MC3T3-E1细胞中CS-PEI的转染效率高于分子量为25K的PEI和Lipofectmine 2000。在体外促骨分化实验中,转染CS-PEI/BMP2后,Realtime-PCR检测MC3T3-E1细胞成骨相关基因的表达均有不同程度上调 ,钙结节的形成量也明显多于对照组,Westernblot结果显示,转染CS-PEI/BMP2后,Hela细胞中磷酸化SMAD-1/5/8的表达明显增加;将CS-PEI/BMP2复合物与明胶海绵支架复合后移植到大鼠颅骨缺损内,Micro-CT结果显示CS-PEI/BMP2组的大鼠颅骨缺损边缘不规则,骨缺损面积明显减小,促骨形成效果优于明胶海绵组,组织学结果显示CS-PEI/BMP2对大鼠各脏器无明显损害。我们的结果表明,CS-PEI纳米共聚物是一种有效的基因载体,其与BMP2基因结合后能够促进骨组织的再生。

Chitosan grafted low molecular weight polyethylenimine deliver bone morphogenetic protein 2 gene to promote bone formation
Abstract:Through combining chitosan (CS) and low molecular weight polyethylenimine (PEI), we achieved a gene vector with high efficiency and low toxicity for delivering bone morphogenetic protein (BMP2) gene in promoting bone formation. The particle size and zeta potential of the PEI-CS/DNA complexs were characterized by dynamic light scattering and stable at 162nm and 26.7mV. With increasing the weight ratio, DNA binding ability of the copolymer increased. CS-PEI copolymer showed excellent biocompatibility in 293T and MC3T3-E1 cells at various concentrations. Compared with PEI 25K and Lipofectmine 2000, the transfection efficiency of CS-PEI was higher in MC3T3-E1 cells and comparable in 293T cells. In order to induce osteogenic differentiation in vitro, BMP2 gene delivered by CS-PEI copolymer showed a stronger effect on MC3T3-E1 preosteoblast cells in terms of the osteogenic related mRNA expression and mineralization compared with the control group. In addition, the secretion and function of BMP-2 were demonstrated via the appearance of phosphorylation of Smad1/5/8 in HELA cells after transfected. In vivo, the CS-PEI/BMP2 complex confirmed its ability to promote bone regeneration and excellent biological safety. Our study suggested that CS-PEI copolymer deliver the BMP-2 gene into MC3T3-E1 cells with highly promoted efficiency and provide a potential gene delivery system for promoting bone regeneration.
