生物钟基因Per1,Per2与Bmal1对小鼠脉管生长的作用

摘要:利用斑马鱼模型进行的研究表明,生物钟基因Per2与Bmal1可参与调节血管发育。小鼠视网膜的血管新生是在出生后开始,而且卵泡发育及黄体形成过程中也伴有周期性的血管生长与退化。本研究结合Per1,Per2以及Bmal1基因敲除小鼠,利用免疫荧光组织化学分析了新生鼠视网膜与皮肤组织、以及成年鼠卵巢与子宫组织中的血管与淋巴管生长,研究发现缺失Per1,Per2或Bmal1基因不影响小鼠卵巢、子宫及其他组织中的脉管发育。生物钟基因在斑马鱼与小鼠脉管生成中的不同作用机制有待进一步研究。

The role of circadian genes Per1, Per2 and Bmall1 in vascular growth
Abstract:Previous studies showed that Per2 and Bmal1 played important roles in the regulation of VEGF-dependent angiogenesis in zebrafish. As we all know, angiogenesis of retina begin after birth. And the process of ovarian cycles is accompanied by the active vascular growth. In this study, we employed genetically modified mouse models including Per1, Per2 single and also Bmal1 null mice, and analysed blood and lymphatic vascular growth in retina and ventral skin of newborn and ovary, uterus at adult stages. We did not observe any obvious difference in both blood and lymphatic vascular growth in ovary, uterus and other organs of mice deficient of Per1, Per2 or Bmal1 compared with the control mice. The different detailed mechanism in vascular growth remains further investigation between zebrafish and mouse
