基于Fe3O4磁性纳米材料及纳米金标记的沙门氏菌可视化检测新方法

摘要:本文建立了一种基于Fe3O4磁性纳米材料,利用纳米金变色效应对沙门氏菌检测的新方法。选择沙门氏菌特异识别寡核苷酸序列的互补配对碱基序列,分别修饰于Fe3O4磁性纳米材料及纳米金表面,构建捕获探针和显示探针,当在体系中加入沙门氏菌目标DNA链后,根据碱基互补配对原则,会形成独特的"三明治夹心"结构。由于纳米金的颜色会随着金颗粒聚集程度的不同发生变化,当加入的目标物质增多,纳米金发生团聚,颜色会相应出现由酒红色至蓝色不等的颜色变化,从而实现可视化检测。在最优实验条件下,本方法在检测沙门氏菌目标DNA时,浓度在1 pmol/L~1500 pmol/L范围内呈现出良好线性关系,线性回归方程为y=0.007x+0.2535(R2=0.9983),最低检出限为1 pmol/L;在检测沙门氏菌时,线性范围为30~8600 cfu/mL,线性回归方程为y=0.0001x+0.3083(R2=0.9962),最低检测限为23 cfu/mL。经特异性和加标回收实验证明本方法可用于实际样品的检测。

A novel method for visual detection of Salmonella based on Fe3O4 magnetic beads and AuNPs
Abstract:In this paper, we established a novel method for visual detection of Salmonella based on Fe3O4 magnetic beads and colorimetric effect of AuNPs. We chose the complementary oligonucleotide sequences of bases which Salmonella can recognized specifically, then modified the surfaces of Fe3O4 magnetic beads and AuNPs with them. So that, we've constructed the capture probe and the display probe. It would form a special "sandwich" structure at the time we added Salmonella target DNA into the system according to the principle of complementary base pairing. As the color of AuNPs changes according to the degree of aggregation of AuNPs, if we added more target DNA, more and more AuNPs aggregated, the color of AuNPs would vary from red to blue. So that we realized the visual detection of Salmonella. Under the optimal conditions, it showed a good linear relationship when the concentrations of target DNA was in the range of 1 pmol/L to 1500 pmol/L, the linear regression equation was y=0.007x+0.2535 (R2=0.9983), the lowest detection limit was 1 pmol/L. And it showed a good linear relationship when the concentrations of Salmonella was in the range of 30 to 8600 cfu/mL, the linear regression equation was y=0.0001x+0.3083 (R2=0.9962), the lowest detection limit was 23 cfu/mL. The specificity and the spiked recovery experiments proved that this method could be used for the detection of actual samples.
