外源金属硫蛋白对热应激下奶牛淋巴细胞活性和抗氧化性能的影响

摘要:为了研究外源性MT对热应激下体外培养奶牛淋巴细胞活性和抗氧化性能的影响。将试验分为4组,每组细胞培养液中MT的终浓度分别为0 mg/ml(对照组)、70 μg/ml(第I组)、140 μg/ml(第II组)、210 μg/ml(第III组),置于培养箱(37℃、5% CO2)中培养,培养20 h后,将其置于43 ℃水浴锅中进行热应激处理1h,在细胞培养箱中培养3 h后,进行各项指标的检测。结果为:1)第I-III组的淋巴细胞活性均显著高于对照组(P<0.01),第I组活性最高。第I-III组的活性呈下降趋势;2)第I-III组的CAT活性、SOD活性和GSH-Px活性均高于对照组,MDA含量均低于对照组。表明MT能够改善热应激下淋巴细胞的活性和抗氧化性能,且浓度为70 μg/ml效果最好。

Effects of Exogenous Metallothionein on Cell Activity and Anti-oxidation of Lymphocytes of Dairy Cattle under Heat Stress
Abstract: In order to investigate influence of exogenous metallothionein on cell activity and anti-oxidation of lymphocytes of dairy cattle under heat stress. The trial was divided 4 groups, and the final concentration of MT in each groups(control group , group I to III) were 0 μg/ml,70 μg/ml,140μg/ml,210μg/ml. Then the cells were cultured in cell incubator (37℃,5% CO2).After 20 h, the cells were cultured in water bath pot(43℃) for 1h. Last the various of indexes were detection. The results as follow: 1) The cell activity of lymphocytes of group I-III were significantly higher than control group and the group I was the highest, but cell activity of was group I-III on downtrend .2) The activity of CAT,SOD,GSH-Px in lymphocytes of group I-III were significantly higher than control group, the content of MDA was lower than control group. According to the consequence , The cell activity and anti-oxidation of lymphocytes were improved by MT and effect of 70μg/mlwas the best.
