维生素E自微乳化载药体系制备及其性质研究

摘要:本文比较了Tween系列、Cremophor EL和Cremophor RH 4等表面活性剂形成维生素E自微乳化载药体系(VE-SMEEDS)的微乳相图。研究了Km值(表面活性剂/助表面活性剂的质量比)对微乳相图的影响。考察了助表面活性剂和表面活性剂对微乳粒径的影响。采用电导率法对VE-SMEEDS稀释过程进行了相区划分。考察了温度、氯化钠和蔗糖浓度对VE-SMEEDS微乳稳定性的影响。结果表明,最佳VE-SMEEDS配方是以Cremophor RH 40为表面活性剂,乙醇为助表面活性剂,Km=3,其中Cremophor RH 40和维生素E的质量比4:3,VE载量为36%。其O/W型微乳的平均粒径为46.32±1.77nm且对温度、NaCl和蔗糖稳定。

Preparation and Characterization of vitamin E SMEDDS
Abstract:This study investigated the pseudoternary phase diagrams of VE self-microemulsifying drug delivery systems(VE-SMEDDS) formed by using different surfactants including Tween series, Cremophor EL and Cremophor RH 40. The effects of different Km on the pseudoternary phase diagrams of VE-SMEEDS as well as the influences of surfactant and co-surfactant on particle size of O/W microemulsion were studied. The phase transition of VE-SMEEDS in the dilution progress was determined by conductance measurement. The stability of VE-SMEEDS as functions of temperature, NaCl and sucrose concentration was also investigated. The results showed that the optimal VE-SMEEDS formulation was obtained when Cremophor RH 40 and ethanol were used as surfactant and co-surfactant respectively (Km=3). The mass ratio of Cremophor RH 40 and VE was 4:3 and the content of VE in the system was 36% (w/w). The average particle size of O/W microemulsion was 46.32 ± 1.77nm. The VE-SMEEDS exhibited good stability on temperature, NaCl and sucrose solution.
