离子液体在生物大分子研究中的进展

摘要:离子液体以其熔点低、蒸气压小、酸碱性可调、良好的热稳定性及溶解性能、粘度、密度等优异的理化特性成为溶解蛋白质、纤维素等生物大分子的新型的绿色溶剂。离子液体在生物大分子中的应用已引起广泛关注。本文综述了生物大分子蛋白质在离子液体中的溶解、结晶行为及离子液体对蛋白质活性及稳定性的影响。相关研究表明,离子液体+水体系可以提供蛋白质溶解所需的氢键环境,提高蛋白质的溶解性。在离子液体中进行酶催化反应,酶的活性一般相当或高于其在常用有机溶剂中的活性,而且酶的热稳定性、操作稳定性和选择性均有提高。通过添加离子液体诱导蛋白质结晶,可以提高蛋白质晶体的X射线衍射分辨率。此外,对纤维素在离子液体中的溶解行为也进行了介绍。

Research Progress of Ionic Liquids in biological macromolecules
Abstract:Ionic liquid is a new developed green solvent system for dissolving bio-macromolecule, such as protein and cellulose. Ionic liquid has intrinsically useful properties,such as good thermal stability, low melting point, negligible vapour pressure, changeable acidity, high dissolvability, low viscosity and density. The application of ionic liquid in macromolecules research has received great attention. The dissolution and crystallization behavior of protein and and the effect ionic liquids on the activity and stability of protein were summerized in this paper. The related research results indicated that ionic liquid + water system could provide hydrogen-band environment for disslution of protein, which can improve disslution behavior. The activity of enzyme is generally equivalent or higher than that of usual organic solvents and the thermal stability, operating stability and selectivity of enzyme are promoted. By adding ionic liquid to induce protein crystallization, X-ray diffraction resolution of protein could be greatly improved. Furthermore,the dissolution behavior of cellulose was also introduced in this paper.
