球孢白僵菌线粒体全序列测定及序列分析

摘要:为了准确地了解球孢白僵菌的核外遗传物质的变异情况,本文进行了球孢白僵菌线粒体基因组全序列的测定。结果获得一个包含有29961个碱基的环状结构,其AT含量为72.75%,该序列公布在EMBL数据库(登录号为EU371503)。该基因组编码了核糖体大亚基(rnl)、核糖体小亚基(rns)、25个与翻译相关tRNA基因和14个参与氧化磷酸化途径的蛋白基因,即7个NADH脱氢酶亚基(nad1- nad6 和 nad4L),3个细胞色素氧化酶(cox1, cox2, cox3),细胞色素b (cob) 和3个ATP合成酶 (atp6, atp8, atp9)。所有编码基因均由mtDNA分子同一条链编码,与粪壳菌纲的其它种相似,其tRNA集中分布在三个簇。与NCBI数据库比对和进化树分析表明,球孢白僵菌能与其它13个真菌可构建成单一进化树,与蝇蚧霉相似率最高,达73%,支持率达100%。

Sequencing and analysis of complete mitochondrial genome of Beauveria bassiana, an important entomopathogenic fungus
Abstract:To exactly understand the extranuclear variation of B. bassiana, the whole sequence was sequenced in the present study. A circular molecule of 29961bp was obtained with an overall A+T content at 72.75%,which was registered in EMBL with the accession number EU371503. It contained the genes encoding for the large and small subunits of ribosome (rnl and rns), 25 genes encoding tRNA and 14 protein encoding genes involved in the oxidative phosphorylation and ATP synthesis, namely genes encoding for three ATP synthase subunits 6, 8 and 9 (atp6, atp8, atp9), three cytochrome oxidase subunits I, II, III (cox1, cox2, cox3), apocytochrome b (cob), and 7 NADH dehydrogenase subunits 1-6 and 4L (nad1, nad2, nad3, nad4, nad5, nad6, nad4L). All genes encoded in the mtDNA genome of B. bassiana were from the same strand and most of the tRNAs arranged in three conserve trn gene clusters which is similar to other Sordariomycetes.. The sequence is higher similar to Lecanicillium muscarium at 73% through alignments with NCBI and phylogenetic analysis with 14 protein-coding genes.
