不同中药对小鼠乳腺β-酪蛋白表达的影响

摘要:中草药主要以自然界的植物,动物和矿物为原料,是动物体最易吸收的纯天然物质,对奶牛效果良好,毒副作用甚微,几乎无残留、无抗药性、不污染环境。因此,根据传统的中兽医理论和现代的中药研究成果,以源自天然的毒副作用小、无残留、不易产生耐药性的中药为原料,可制成无抗生素的复合型绿色饲料添加剂。奶牛添加剂的研究与应用受到广泛重视。许多具有补益气血、通经活络、调和脾胃的中草药及其复方制剂均具有显著的催乳作用。本试验在分子水平上进行了如下的研究。在人下奶方中草药复方中,选取5种广泛应用的中草药:王不留行、甲珠、通草、蒲公英、漏芦。通过普通的中草药煎煮方法制成一定比例的药液,经过过滤制成无菌药液。取哺乳5~12d昆明小鼠乳腺上皮细胞作为研究对象。将上述5种中药液分别加入细胞培养液中,并与空白组和加催乳素组进表达β-酪蛋白的情况进行多种的比较。运用MTT法测定上述5种中药对体外小鼠乳腺上皮细胞增殖的影响;运用激光共聚焦显微镜比较周期蛋白D1和Stat 5;运用荧光定量RT-PCR方法定量检测β-酪蛋白基因;运用毛细管电泳仪检测β-酪蛋白;将药液直接注射到哺乳期中小鼠乳腺中测产奶量。实验结果表明,甲珠、通草和漏芦对小鼠乳腺上皮细胞的增殖有明显的促进作用,而王不留行对小鼠乳腺上皮细胞的增殖有极其明显的促进作用。蒲公英对小鼠乳腺上皮细胞的增殖无明显作用。

Effect of expression of β-Casein on mice\\\
Abstract:Chinese herbs,which is made up by plants,animals and mineral,is a natural resource can be absorbed most easily by animals. And Chinese herbs ,which has less side- effect,less remains,less drug-resistance and leads less pollutions,causes better effects on dairy cows. Thus,people use Chinese herb as raw material to product the mixed additive including no antibiotics. Research and use of the additive on milk cows are the focus of current scholarly pursuit. The study,as following ,is on the level of Biochemistry and Molecular biology. We selected five Chinese herbs among the compound recipes extensively used in human beings to increase the level of milk ,they are Cowherb Seed,JiaZhu,Rhaponticum uniflorum,Medulla Tetrapanacis and Taraxacu Mogono. The herbs being boiled,we extracted the juice that includes no bacterium.The research use the cells of mammary gland of mouse that feeds its babies more than 5 days and less than 12 days. We added the herbs into the culture medium of cell respectively and did a rang of comparisons –the expression of β-Casein compared-with the circumstances without additive and only within prolactin. There are five parts in the experiment .First,we use the approach of MTT to evaluate how the herbs influence the cells on cell proliferation. Second,we studied the regulation of cyclin D1 and Stat 5 by laser scanning confocal influorescence microscopy. Third,Real time Quantitative PCR. Forth,capillary electrophoresis is used on detecting β-Casein ,too. Fifth,injecting the juice into the mammary gland of mouse on lactation to measure quantity of milk.The result indicated that the pangolin scales ,medulla tetrapanacis and rhapontici can restrain the multiplication of mouse mammary epithelial cells(P<0.05),and cowherb seed can restrain the multiplication of mouse mammary epithelial cells extremely(P<0.01). But,dandelion can not restrain the multiplication of mouse mammary epithelial cells.
