柽柳谷胱甘肽S-转移酶基因(ThGST1)克隆与分析

摘要:本研究运用PCR技术,从刚毛柽柳cDNA文库中分离出谷胱甘肽S-转移酶基因cDNA,命名为ThGST1。序列分析表明,ThGST1基因具有726 bp开放读码框,编码含241个氨基酸的蛋白质。推测编码蛋白的分子量为27.47 kDa,理论等电点(pI)为9.61。采用实时荧光定量RT-PCR]方法研究了PEG和NaCl胁迫后刚毛柽柳不同组织中ThGST1基因的表达模式。结果表明: PEG和NaCl处理均能诱导柽柳根中ThGST1 基因的表达。

Cloning and expression of a glutathione S-transferase gene from Tamarix hispida .
Abstract:The full-length cDNA of a presumed glutathione S-transferase (ThGST1) gene was isolated from a cDNA library of Tamarix hispida leaves. The ThGST1 gene contains 726-base pair (bp) open reading frame (ORF) and encodes a 241-amino-acid polypeptide with a predicted molecular weight of 27.47 kDa and pI of 9.61. Expression of ThGST1 in roots and leaves of T.hispida with PEG and NaCl treatments was studied using real-time RT-PCR. The results showed that ThGST1 transcripts were induced by PEG and NaCl stresses in the roots.
